I am currently taking a public health course where we recently examined the devastating effects of environmental contaminants such as arsenic and lead, from the Flint, Michigan water crisis to arsenic exposure through private well water in Maine. Learning about the biochemical explanations behind these public health issues was especially engaging.

The lipoamide arm contains sulfhydryl (-SH) groups that are essential for transferring acetyl groups to coenzyme A. When arsenite binds to these -SH groups, it locks the lipoamide in an inactive form, halting the enzyme’s catalytic cycle and blocking the production of acetyl-CoA. This disruption impairs cellular respiration and helps explain the severe metabolic consequences observed in arsenic poisoning.

I am excited to bring this biochemical understanding into my public health class discussions and to find ways to connect it with my thesis research on PFAS contamination in Maine. Examining how environmental toxins disrupt molecular pathways has deepened my appreciation for the connection between biochemistry and environmental health and strengthened my motivation to understand these mechanisms as a foundation for addressing contamination and its impact on human health.

In Monday’s discussion of the citric acid cycle, I was particularly interested in the role of the lipoamide “swinging arm” in the pyruvate dehydrogenase complex.
